بیماری شناسی گیاهی دانشگاه تربیت مدرس

دهه گذشته تغیرات اساسی در دانش ما در مورد سفیدک های پودری (Erysiphales) ایجاد شده است. تحقیقات روي فیلوژنی مولکولی نشان داده است که سفيدكهاي پودري يا Erysiphalesجزء لئوشیومایست (Leotiomycetes (دیسکومیست های بدون اوپرکول) می باشند در صورتی که قبلا جزء پیرنومیست ها یا پلکتومیست ها در نظر گرفته می شدند . چرخه زندگی سفیدک ها به طور قابل توجهي متغیر می باشد و داراي هر دو مرحله جنسی و غیر جنسی یا فقط مرحله جنسی یا فقط مرحله غیر جنسی می باشد. حداقل یک گونه، کنیدی های رنگی ( (Dematiaceous conidia تولید می کند. بررسي توالی های DNAي ریبوزومی نشان می دهد که تمایز تاکسون ها در آنها بیشتر مربوط به خصوصیات غیر جنسی مانند اونتوژنی و شكل کنیدیها (در مقابل خصوصیات جنسی) می باشد. استفاده از مدل های ساعت مولکولی محققان را قادر ساخته است که الگوهای تکامل همزمان (coevoloution) و پرش میزبانی (Host-jumping) و نيز الگو های مهاجرت باستانی را بازسازی نمایند. به نظر می رسد پراکنش جغرافیایی برخی گونه ها به سرعت در حال افزایش باشد اما در مورد تنوع گونه ها در مناطق وسيعي مانند آمریکای شمالی اطلاعات کمی وجود دارد. این موضوع که احتمالاً سفیدک های پودری قبلاً تحت تاثیر تغیر آب و هوا بوده‌اند ، نشان مي‌دهد که آنها مي‌توانند مدل های مفیدی برای مطالعه ی اثرات تغییر آب و هوا روی بیماری های گیاهی باشند.

RNA Miniprep from Neurospora (Fungal Genetics Stock Center)
Procedures of RNA extraction published so far follow roughly three different approaches, where phenol/chloroform, guanidinium salts, and/or LiCl is used. However, these protocols have some disadvantages. This method, combining the advantages of phenol extraction and LiCl precipitation, is for isolating high quality total RNA from N. crassa mycelia that reliably yields large quantities.

Most methods of DNA preparation from fungi are time-consuming due to the need to first make protoplasts, expensive for chemicals such as cesium chloride, or suitable only for small scale preparations.
